The performance, in Los Angeles on February 24, will be the first time she has sung the song live on stage.
Skyfall is in the running for the best original song award at this year's Oscars and is the first Bond song nominated since For Your Eyes Only in 1981.
She said: "It's an honour to be nominated and terrifyingly wonderful to be singing in front of people who have captured my imagination over and over again.
"It's something I've never experienced and probably only ever will once."
The singer - who attended the ceremony with Robbie Williams' wife Ayda Field - has kept a low profile since she gave birth in October. She and boyfriend Simon Konecki have still to reveal the name of their son, although she took the opportunity to dedicate the award to him.
As she collected her award, Adele gushed: "Oh my God - honestly, I came out for a night out with my friend Ayda - we're new mums. We've literally come for a night out.
"I was not expecting this. It's very strange to be here - thank you so much for letting me be a part of your world for a night.
She went on: "And this is for my boyfriend Simon who convinced me to do it and for my lovely son - thank you so much."
