The pair have signed up to the Extremely Loud And Incredibly Close director's big-screen adaptation of Andy Mulligan's 2010 novel of the same name, which has been adapted for the screen by Richard Curtis, reported Deadline.
The West Wing actor has been tipped to play Father Juilliard, while Rooney is expected to portray a charity worker.
Newcomers Rickson Tevez, Eduardo Luis and Gabriel Weinstein are set to play three poverty-stricken boys who discover something unusual, mysterious and dangerous in a city rubbish tip.
Production is expected to start in Rio de Janeiro in August, in time for a release in summer 2014.
Rooney, last seen in Steven Soderbergh's thriller Side Effects, next stars in Spike Jonze's drama Her alongside Joaquin Phoenix and Olivia Wilde, along with Terrence Malick's as-yet-untitled film. She is also tipped to reprise her role as talented hacker Lisbeth Salander in the next instalment US remake of Stieg Larsson's Millennium trilogy, The Girl Who Played With Fire.
Martin, best known for starring in war drama Apocalypse Now, Wall Street and The Departed, will reprise his role as Peter Parker's uncle Ben in The Amazing Spider-Man 2.
