Anna was set to star in Zach Braff's directorial debut Wish I Was Here, but scheduling conflicts meant that Ashley has now taken over her role, reported The Wrap.
Zach, whose movie was partially funded through crowd-funding website Kickstarter, announced the casting change in a message to campaign backers.
He said: "Well, since you're having your first shot as being movie producer, it's time you experience something that happens a lot: scheduling conflicts.
"We've tried our hardest, but we've been unable to figure out the lovely Anna Kendrick's scheduling conflicts with another movie she's doing. This happens all the time in movie-land. Putting the shooting schedule together is like planning 25 weddings: each day has so many moving pieces and locations. I love Anna so much and can't wait to work with her one day."
But Zach said he was thrilled to have Ashley on board, referring to her as "charming" and "hilarious".
He later tweeted: "So excited that @AshleyMGreene has joined the cast of Wish I Was Here. Almost time to start..."
The film will see Zach play a struggling actor, father and husband in his 30s who reconnects with his family as he searches for a purpose in life. Kate Hudson, Josh Gad, Mandy Patinkin and Jim Parsons have also joined the cast.
Ashley played vampire Alice Cullen in the Twilight movies, while Anna starred as human Jessica Stanley.
